Applications of BBa_K2287001

In 2017, team UCAS used this part to compose a ammonia sensor. The working circuit is registered as BBa_K2287014. Click here for more imformation.

Following are characterization of the promoter which reveals that this part is a reliable promoter for engineering.

Figure 1: Delay time of promoter glnAp2 under different concentrations of ammonium. Abscissa are relative concentration of ammonium.

This part reveals predictable behavior and high resolution to nitrogen concentration (fig.1). On the one hand, the concentration of nitrogen and the response time shows a obvious linear relationship. On the other hand, the high signal-noise ratio implies that the sensor is usable even when ammonium reaches extremely low level.

Figure 2: Kinetic curves of glnAp2 over time of glnAp2 under different concentrations of potassium nitrate. The strength is represented by the quotient of fluorescence intensity over OD 600.

Kinetic curves shows that nitrate does interfere this part detecting ammonium (fig.2). However, this interfere is quite weak when the concentration of nitrate is low, which can be ignored in application.

The working circuit consists of the promoter glnAp2, the reporter mRFP1 and the moderate degradation tag DAS. Kinetic curves show that
